Anyone know why this wheel has a direction?....

On an F1 car, aerodynamics are more important. Diverting airflow laterally through the wheels is very inefficient when you're looking for absolute airflow efficiency, so the brakes are cooled via ducted airflow taken from just the right place to provide the correct characteristics for a given car. No need for cooling via the wheels.

 

For this reason they always pop a blower onto the cooling ducts whenever a car comes back into the pits to stop the brakes from overheating.

Same as the Lexus ISF, each wheel has its own corner due to staggered width and blade direction. You often see them incorrectly fitted with the thicker section of the blades trailing when they should be leading (with the wheel rotating clockwise, of course),

Hasn't the ISF also got ducting off the grill to focus cooling.... Very few OEM cars today have handed wheels, only Lancia and Lambo come to mind.

Yes it has, also vents in the front wings to expel the air from the front ducts.
